To isolate protein, fraction techniques are used. This is a conventional way of purifying proteins, which includes the proteins’ solubility, size, charge, and bending specificity . To begin protein purification, the sample must be isolated from its environment. To carry on this task, the cells containing the protein of interest must be harvested and ruptured, which can be done by centrifugation.
Cells contain thousands of proteins of varying function and characteristics so knowing the desired protein’s properties is key as the composition of proteins determines how they are separated and what tools and reagents are necessary to isolate them. For example, if our protein of interest consists of amino acids that are positively charged, the beads used in the Ion-exchange technique must contain negatively charged beads in order for the positively charged proteins to be “captured” by forming a bond with the beads and allowing for the negatively charged proteins to be separated.
